Spermula is a fantasy film with erotic scenes that tells the story of a post-apocalyptic sect seeking the means to procreate. The film opens with a description of a group of people who were sexually adventurous and socially progressive. They associated sex with total freedom and existential autonomy and were composed of unusual individuals from all walks of society. When a conference celebrating their vision is arranged in New York in 1937, the sect suddenly vanishes.

Years later, an enterprising young reporter attempts to locate the sect and travels through the Amazonian rainforest following leads to their whereabouts. A title card emerges at this point in the film: "This film is the story of some of their daughters." A giant boat with winged extensions is seen traveling through the sky. A woman and her colleagues emerge. She is named Spermula and she seeks to return to the world to deliver a message of enlightenment.

Spermula seeks to acquire massive quantities of semen from men so as to rid human males of their aggression, possessiveness, territoriality, and proclivity for violence. She and her friends live in an old, ruined estate in a small town. The mayor is an abusive monster and his assistant is engaged in an incestuous relationship with her son.

Spermula decides to initiate a massive group orgy with all the people in the town, but her plot fails. Her colleagues have been derailed by the attractions and poisons of earth and Spermula herself forfeits her immortality for a one-night stand with an art student. Later, a planet, also named Spermula is inhabited solely by women who seek to emasculate men by vacuuming out their semen. For the latter half of the film, the natives of Spermula, like Spermula herself, forfeit their relatively cozy existence for sexual intercourse with human males. One man, Werner, becomes a teacher after being transformed into a man from his previous feminine state and has sex with every Spermulite.
