William "Uncle Bill" Davis (Brian Keith), a bachelor engineer who now works at a construction company, becomes the legal guardian of his orphaned nieces and nephew: Cissy (Kathy Garver), Jody (Johnny Whitaker), and Buffy (Anissa Jones). In the pilot episode of Family Affair, Buffy, Uncle Bill's cousin Enid is on a six-month European vacation. In the meantime, she entrusts her children, Cissy, Jody, and Buffy, to Uncle Bill's care.

Buffy is the youngest of the three children and the center of attention in this debut episode. Uncle Bill, who is used to his quiet life, is apprehensive about having to take care of three young children, but he gradually gets used to it.

Buffy is an adorable and inquisitive little girl who is curious about everything around her. Cissy and Jody are protective of Buffy and her needs, but they have their own agendas that they want Uncle Bill to comply with.

While on a shopping trip with Uncle Bill, Buffy becomes fascinated by the toy department, particularly by a doll she wants. Uncle Bill initially refuses to buy it, but Buffy throws a tantrum, and he gives in.

While she is admiring her new doll at home, Buffy overhears Uncle Bill tell Cissy and Jody that they have to go to Washington, D.C., for a few days. This news upsets Buffy, and she insists that she stays behind. She promises that she will be a good girl and look after herself, but Uncle Bill is having none of it.

The children eventually convince Uncle Bill to take them to Washington, D.C., as it is a trip they had planned with their parents before their passing. The children are excited by the change of scenery, and Uncle Bill is determined to make sure everyone has a good time. Uncle Bill has a difficult time adjusting to the children's unpredictable behavior, but he manages to have fun with them.

While in Washington, D.C., the family takes in the sights and sounds of the city. They visit the White House, the Lincoln Memorial, and the Smithsonian Museum. The family also meets a new friend, an African American chef named Frenchy (Sebastian Cabot), who works in a restaurant and offers to cook for the children. Uncle Bill and Frenchy's newfound friendship sparks a new adventure for the family.
